Quick answer: Lift the tail and look at the two openings. A male has a round opening about half an inch below the anus; a female has a vertical slit almost touching it. Compare same-size littermates when you can.
A male kitten’s two openings sit about ½ inch apart; a female’s slit sits almost against the anus. That spacing-and-shape check is the published cue that answers the query on its own. The Three-Check Sexing Rule
Our research team used one selection rule. A cue had to name both opening shape and the gap from anus to genitals. Face-shape myths, personality guesses, and any kit sold as a “gender test” stayed out.
We call the keepers the Three-Check Sexing Rule. Check distance first. Check shape second, then age, because tissue fills in as the kitten grows.
Humane World for Animals, on its HumanePro fact sheet, describes male spacing as relatively wide, about ½ inch. It describes a female’s anus and vaginal opening as close together, almost touching.
The ASPCA’s kitten-sexing sheet uses the same pair of cues without a tape measure. It calls the female opening a vertical slit set directly below the anus. It calls the male opening a small round dot set a greater distance away, with the testicles between.
Washington State University’s College of Veterinary Medicine repeats that pair. The genital opening is round in males and a vertical slit in females. In kittens of similar size, the anus-to-genital gap is greater in the male.

How to lift the tail without chilling the kitten
Sexing is a look, not a workout. Rest the kitten on a warm towel, belly down. Lift the tail only high enough to see the perineum.
Best Friends Animal Society tells bottle-baby caregivers that a kitten’s ideal body temperature is 100 to 102 degrees Fahrenheit. It also says never to feed a cold kitten. Keep handling short, then return the kitten to a covered heat source.
Best Friends also says bottle babies need a heating pad until they are 3 to 4 weeks old. Leave room for the kitten to crawl off the warm spot.
Do not scrub the area with adult dog shampoo to part the fur. Those formulas belong in our guide to dog shampoos for itchy skin, which is written for dogs. Neonatal skin is not that audience.
If fur hides the openings, use a warm, damp cloth. Best Friends tells caregivers not to put soap or pet shampoo directly on a young kitten.

What the two openings look like
Start at the base of the tail. The first opening is the anus. It is round in both sexes.
The second opening is the one that tells you the sex.
In a female, Humane World for Animals says the vaginal opening looks more like a vertical slit or a teardrop than a round hole. The two openings sit almost together.
Many caregivers remember that pair as an upside-down exclamation mark. The period is the anus. The line is the slit.
In a male, Humane World for Animals says the area around the hidden penis looks more like a circle than a slit. The gap is the space where the scrotum will sit.
Many caregivers remember that pair as a colon. Two dots. A gap between them.
The ASPCA notes that the same distance guide also works on adult cats.
If you have a litter, do the same lift on each kitten. The ASPCA says comparing littermates is particularly helpful. Washington State University makes the same point for kittens of similar size.
One kitten alone can look ambiguous. Two kittens side by side make the gap obvious.
Do not press hard. Humane World for Animals says you may gently palpate a suspected male’s scrotum with thumb and forefinger. Two small oval objects under the skin point to a boy.
Skip that step if the kitten is distressed. Distance and shape still stand.
Age and weight change what you can see
The anatomy is present from birth. The view is not equally clear at every week.
Humane World for Animals says testicles will not be plainly visible until the kitten reaches 6 to 10 weeks of age. You should still see extra fur and tissue in the gap before that.
Washington State University’s teaching photos are labeled as 8-week-old kittens. By then, the male’s testicles are becoming more apparent.
Weight helps you guess age when the birthday is unknown. The ASPCA’s 2023 Kitten Age & Weight Chart lists a day-one range of 70 to 115 grams, with an average near 100 grams. The same chart says normal kittens gain 7 to 15 grams per day.
Use those ranges with developmental milestones, not as a lone clock.
Best Friends Animal Society lists a birth weight of about 2 to 4 ounces. It says kittens should gain about ½ ounce every day, or 3 to 4 ounces per week. By 8 weeks, most kittens weigh about 2 pounds.
Eyes start to open at 7 to 10 days. They are fully open by 20 days.
PetAg’s KMR Kitten Milk Replacer Powder page is useful when you are sexing a bottle baby you also have to feed. PetAg tells caregivers to mix 1 part powder with 2 parts warm water.
It tells them to feed 2 tablespoons, or 30 mL, of reconstituted formula per 4 ounces, or 115 grams, of body weight each day. It splits that daily amount across feedings every 3 to 4 hours for kittens from newborn to 6 weeks.
PetAg lists a calculated calorie content of 29.6 kcal per tablespoon of powder, or 12.4 kcal per tablespoon once mixed 1:2. The guaranteed analysis on that page is a minimum of 40.0 percent crude protein and 28.0 percent crude fat.
Those numbers do not sex a kitten. They keep a small kitten growing until the 6-to-10-week view is clearer.
Cornell University’s College of Veterinary Medicine, through the Cornell Feline Health Center, says dry cat food contains between 6 and 10 percent water. It says canned food has a moisture content of at least 75 percent. It also says treats should not exceed 10 to 15 percent of daily calories.
Cornell names the Association of American Feed Control Officials as the body whose Feline Nutrition Expert Subcommittee sets basic minimums. Look for an AAFCO nutritional adequacy statement on any kitten food you use after weaning.
Coat color is a clue, not a verdict
Color can support the tail check. It cannot replace it.
Washington State University says almost all, but not all, calico or tortoiseshell kittens are female. It also says more orange kittens are male than female, though that link is weaker than the calico link.
The Animal Medical Center in New York explains why. The gene for orange sits on the X chromosome. A typical male has one X, so he is orange or not orange.
A typical female has two X chromosomes, so she can show orange and black at once. That is the calico or tortoiseshell pattern.
The Animal Medical Center also publishes a hard number. Only 1 in 3,000 male cats are calico. Those males have two X chromosomes and one Y chromosome.
The University of California, Davis Veterinary Genetics Laboratory uses the calico pattern to teach X-inactivation. One X is randomly turned off in each cell early in development.
Orange patches silenced the black recipe. Black patches silenced the orange recipe. That is why two genetically identical calicos can still look different.
It is also why a tricolor coat is a female clue, not a female proof.
If the kitten is solid gray, tabby, or black, color tells you nothing. Go back to the tail.

Can I tell sex on the day a kitten is born?
Yes, the openings are already there. The ASPCA says the job is simply harder in the first few days. Use shape and spacing, not testicles, then put the kitten back on warmth.
When can I feel testicles?
Humane World for Animals says they will not be plainly visible until 6 to 10 weeks. You may feel two small ovals sooner if you palpate gently. If you cannot feel them, trust the wider gap and the round opening.
What if I have only one kitten?
You lose the littermate comparison that both the ASPCA and Washington State University recommend. Use a lighted magnifier and a same-day photo of the perineum. Recheck at 6 to 8 weeks, when Washington State University’s teaching photos show a clearer male scrotum.
Are orange kittens always male?
No. Washington State University says more orange kittens are male than female, but the link is weaker than the calico link. Confirm with the tail.
Can a calico or tortoiseshell kitten be male?
Rarely, yes. The Animal Medical Center puts male calicos at 1 in 3,000 male cats and ties the pattern to an XXY chromosome set. Still lift the tail; a tricolor coat is a strong female clue, not a finished answer.
